Die Handlung von Mitsuboshi Colors findet an einem bestimmten Ort eines bestimmten Parks im Ueno-Gebiet von Tokio statt. An diesem Ort steht die geheime Basis der Gerechtigkeitsorganisation, genannt „Colors“. Die drei Grundschülerinnen Yui, Sacchan und Kotoha führen die „Colors“ an. Die Mitglieder der Gruppe durchstreifen den Park Tag und Nacht (eigentlich nur bis zum Abend), um den Frieden des Parks zu schützen.
Residing within Tokyo's district of Ueno are the Colors, three individuals who protect their city by performing good deeds and aiding their community. Or, at the very least, they pretend to be the city's defenders. In reality, the Colors are just three young girls who spend their time playing make-believe and exploring the city. The Colors' activities are facilitated by the grandfatherly Daigorou "Pops" Kujiraoka, who uses his store's inventory of knick-knacks to entertain the rambunctious trio.
Not everyone is a fan of the Colors though. The local policeman Saitou just wants to deal with his regular duties, but he often finds himself the target of the Colors' attention, having been made the villain in most of their fantasies. But despite his personal feelings, Saitou always finds the time to go along with the three girls' games. Even though the Colors do not actually defend Ueno, they definitely help brighten everyone's day.

Au cœur du parc d’Ueno se trouve un repaire secret entouré de mystères.Trois petites filles s’y retrouvent quotidiennement. Cependant, il ne s’agit pas de fillettes ordinaires mais des membres de l’organisation des justiciers Colors dont la mission est de protéger Ueno.Yui, Sacchan et Kotoha font des rondes jour et nuit pour garder leur ville en paix (enfin...surtout en fin d’après-midi, après la fin des cours).
La storia si svolge in un certo angolo del parco di Ueno a Tokyo. Lì sorge la base segreta di una “Organizzazione per la Giustizia”, chiamata “Colors”.
A capo di questa organizzazione ci sono tre bambine delle elementari, Sacchan, Yui e Kotoha, che assieme agli altri membri sorvegliano il parco giorno e notte (ma in realtà solo fino a sera) per mantenere la pace del luogo. Il loro non è però un compito così facile, visto che si imbattono ogni volta nel loro acerrimo nemico: Saitou, il terribile poliziotto di Ueno.

結衣、さっちゃん、琴葉の3人は今日も平和な上野の平和を守るため、日夜(嘘、夕方まで)街を駆け回るのである!!

A história se passa em um certo canto de um certo parque em uma certa área de Ueno, em Tóquio. Neste parque existe a base secreta da "organização da justiça" chamada "Colors". Um grupo de três garotas da escola primária lidera a "organização", e os membros do grupo vigiam o local dia e noite (na verdade apenas até o anoitecer) para proteger a paz do parque.
В детстве все дела кажутся очень важными, даже самые пустяковые. Но в том и проблема взрослых, что мы не обращаем внимания на мелочи, а ведь они порой перерастают в настоящие трудности, от которых избавиться уже гораздо сложнее, нежели чем пресечь всё на корню. Но жители японского городка Уэно, в отличие от нас, могут спать спокойно, ведь на страже их города стоит настоящая секретная организация "Трёхцветные Звёздочки”, состоящая, как вы уже догадались, из трёх крайне бдительных агентов. Юи, Саччан и Котоха в мирской жизни учатся в начальной школе, но после уроков, и пока родители не позовут домой, они защищают Уэно, неустанно следя за проблемами этого города из своего тайного убежища. Но представитель правоохранительных органов в лице патрульного Сайто никак не хочет сотрудничать с их организацией, а может быть просто завидует, ведь раскрытых дел в копилке девочек куда больше, чем у него.
En un cierto rincón de cierto parque en cierta área de Ueno en Tokio, existe la base secreta de la "organización de justicia" llamada "Colors". Un grupo de tres niñas de escuela primaria dirigen Colors, y los miembros de este grupo deambulan por el parque día y noche (en realidad solo hasta la noche) para proteger la paz del parque.
