In September 1543, Takeda’s army besieges Nagakubo Castle in Shinano. Harunobu decides to give Kansuke the position of Takeda’s military strategist, if he successfully captures the castle. With the help of the spy he has already sent into the enemy, Kansuke swiftly takes the castle. Meanwhile, Yu is in agony, torn apart between her affection and grudge toward Harunobu. Harunobu tells Kansuke to find out Yu’s true feelings.
天文１２年（１５４３）９月。武田勢は信州長窪城を包囲した。勘助（内野聖陽）は力攻めを戒め、策を用いて城を落とそうと謀っていた。家老・諸角虎定（加藤武）が勘助を正式に軍師として用いるよう晴信（市川亀治郎）に進言する。晴信は城攻めが成功すれば勘助を軍師とすると決める。城に籠もっていたのは相木市兵衛（近藤芳正）を始めとする各地で武田に敗れた信州の豪族たちであり、その中に平蔵（佐藤隆太）とヒサ（水川あさみ）の姿もあった。しかし、その相木こそが勘助が城に送り込んだ裏切り者だった。武田勢は相木と呼応して瞬く間に城を落とす。策が当たった勘助はついに武田家軍師として認められた。しかし、武田家には気がかりなことが残っていた。由布姫（柴本幸）が晴信に心底から従わないことであった。晴信に愛情を感じる一方で武田への恨みを捨てきれず姫は苦しんでいた…。
