滿族少女葉赫那拉·玉蘭（劉雪華飾）聰慧志高，不甘於貧窮和男尊女卑的處境。 道光帝六皇子奕訢（伍衛國飾）南下視察民情，邂逅玉蘭於雪地之中，互生情愫。 奕欣又與漢人李樂天（劉緯民飾）結為莫逆之交，李樂天亦鍾情玉蘭。 道光急召奕訢返京，奕訢不及向玉蘭道別而使二人間產生種種誤會，玉蘭決定入宮選秀女。 奕欣在皇位爭奪中落敗，四皇子繼位為咸豐帝，奕欣則封為恭親王，並遭咸豐（黃浩義飾）猜忌和權臣肅順（王偉飾）排擠。 玉蘭入宮受寵，誕下皇子載淳，晉封懿貴妃，逐步展露政治才能和權力野心。 咸豐帝駕崩，留密詔給皇后（麥翠嫻飾）並命肅順輔政，防止懿貴妃奪權。 同治帝登位，咸豐後與懿貴妃分封為東宮太后慈安與西宮太后慈禧。 慈禧得到恭親王和李樂天幫助，從肅順手中奪回權力，展開垂簾聽政，誅殺異己並騙慈安燒毀咸豐密詔。 恭親王對慈禧所為頗心寒，決定辭官回鄉，慈禧得到權力卻失去友情和愛情。
