次世代を担う若手お笑い芸人たちが、有吉弘行が用意した『お笑いの壁』に挑戦し、壁を超え芸人として成長する」というコンセプトの番組。 有吉とアシスタントの進行のもと、話題の次世代お笑い芸人たちがお笑い企画にチャレンジする。また、お笑いの手本『先輩の壁』として中堅芸人も参加する。 番組の最後には最多クリア者が発表され（第6弾まで）、その後有吉の独断と偏見で優勝が発表されるが、優勝がレギュラー参加者への卒業フラグになることもある。
