Lorsque Sam, Clover et Alex, trois adolescentes fraîchement arrivées à Beverly Hills, sont recrutées par le WOOHP - le World Office des Opérations Hautement Prioritaires - pour devenir espionnes internationales, c'est le choc ! Après un sérieux entraînement, les filles sont envoyées par Jerry - leur nouveau mentor - sur leur première mission. Laquelle est de retrouver Peppy Garou, un coach psychologique pour animaux domestiques, mystérieusement disparu de son cabinet de Beverly Hills. Les filles vont bientôt découvrir que Peppy, ainsi que de nombreux de leurs camarades de lycée, ont été enlevés après avoir été «fabulizés". Entendez là, après être rentrés dans une très étrange machine et en être ressortis avec une toute nouvelle allure ! Jonglant entre leur vie quotidienne de lycéenne et leur nouvelle identité secrète, les Spies finissent par démasquer qui se cache derrière ces enlèvements. C'est Fabu, un ancien mannequin déchu et inventeur du «Fabulizeur". Son plan ? Créer un monde idéal peuplé de gens au look fabu-leux sur une station spatiale et détruire la Terre avec tous ceux qui ont refusé sa vision du monde ! Est-ce que l'inventivité, la spontanéité et les gadgets suffiront aux filles pour sauver Beverly Hills et le monde ?

The movie begins with Sam, Alex and Clover starting their new lives in Beverly Hills, California. As each of them were about to cross paths outside a sushi restaurant, nearby of WOOHP agents (including Jerry) purposely cause the giant sushi roll above the entrance to break off and chase after them, setting some nearby animals free in the process. The giant sushi roll, with the three girls log rolling on top of it, chases a baby pig to the street. The three girls are able to avoid it, save the life of the baby pig (which Alex later adopts and names "Oinky") and destroy the sushi roll before it causes any damage. After that, Alex, Sam and Clover introduce themselves and this starts their friendship when they later see each other again at their new school, meet the current principal Miss Skrich and their rivals, Mandy and Dominique and Caitlin. Clover offers her new friends a change of clothes after Mandy sprays them with a chai machine, but they find themselves sucked in through a locker and into one of the offices of WOOHP. Here, they meet Jerry and fellow WOOHP agent Tad. Jerry reveals that WOOHP had been observing the three of them secretly since childhood, showing videos of each girl, and picking them as prime recruits for the organization. However, the girls are quick to reject the invite and refuse to join. However, they later are "forced" into training after each having traumatic experiences that seemingly relate to WOOHP. They agree to go through training and in 48 hours, complete the training.

In dem ersten Film der Spioninnen geht es um den Anfang ihrer Agentenkarriere. Sam, Clover und Alex treffen sich zufällig in Beverly Hills, wo sich zusammen des Angriffs einer Riesen-Sushirolle erwehren müssen. Da es ihnen gelingt, die Attacke dieser agressiven Fisch-Spezialität abzuwehren und sich auch ansonsten gut verstehen, beschliessen sie Freundinnen zu werden, vor allem als sie herausfinden, dass sie alle drei auf die Beverly High School gewechselt sind
